With the biotherapeutic market gearing towards the production of multi-specific antibodies, new challenges await Chinese hamster ovary (CHO)-based cell line development. For bispecific antibodies (BsAbs), high levels of productivity are often driven by optimized vector designs that achieve delicately balanced chain expression. Additionally, consideration of product purity earlier in the upstream process is paramount since product related impurities are common with these complex proteins. This study investigates the development of a holistic CHO cell line development strategy tailored to a range of BsAbs including vector development, transfection screens for optimal stable pool generation, high value analytics including high throughput purity assays and mass spectrometry, and downstream purification process optimization workflows based on molecule type.
